چکیده

Histone acetyltransferases (HATs) are a class of epigenetic enzymes involved in critical cellular processes like nucleosome assembly, DNA damage repair, and transcriptional regulation. HATs are implicated in many human pathologies including cancers. This chapter describes essential experimental considerations for performing high-throughput screening assays, and offers practical strategies for assay optimization and validation. Illustrative walkthroughs for several HAT assay formats are provided. This content should be useful for those performing HTS assays, orthogonal assays, and counter-screens involving HATs in the context of drug discovery, chemical biology, and molecular pharmacology. 1 Division of Pre-Clinical Innovation, National Center for Advancing Translational Sciences, National Institutes of Health, Rockville, MD, USA. 2 Chemical Biology Laboratory, Center for Cancer Research, National Cancer Institute, National Institutes of Health, Frederick, MD, USA. 3 Quantitative Biology, Eli Lilly and Company, Indianapolis, IN, USA. 4 Department of Pathology, Brigham and Women’s Hospital, Boston, MA, USA; Email: [email protected]. Nucleic acid-mimicking coordination polymer for label-free fluorescent activity assay of histone acetyltransferases.

A novel and label-free fluorescence assay for histone acetyltransferase (HAT) activity was established via in situ generation of a nucleic acid-mimicking CoA-Au(I) coordination polymer (CP). Moreover, the potency of this assay for HAT-targeted drug discovery was proved by screening HAT inhibitors.

Epigenetic targets and drug discovery Part 2: Histone demethylation and DNA methylation.

Chromatin structure is dynamically modulated by various chromatin modifications, such as histone/DNA methylation and demethylation. We have reviewed histone methyltransferases and methyllysine binders in terms of small molecule screening and drug discovery in the first part of this review series. In this part, we will summarize recent progress in chemical probe and drug discovery of histone dem...

A duplexed phenotypic screen for the simultaneous detection of inhibitors of the molecular chaperone heat shock protein 90 and modulators of cellular acetylation.

Histone deacetylases (HDACs), histone acetyltransferases (HATs), and the molecular chaperone heat shock protein 90 (HSP90) are attractive anticancer drug targets. High-throughput screening plays a pivotal role in modern molecular mechanism-based drug discovery. Cell-based screens are particularly useful in that they identify compounds that are permeable and active against the selected target or...

A long-lived luminescent probe to sensitively detect arylamine N-acetyltransferase (NAT) activity of cells.

Arylamine N-acetyltransferase (NAT) is an important phase II metabolizing enzyme that influences drug efficacy and adverse effects. Here, we report a long-lived luminescent lanthanide complex as a probe for NAT, employing an intraligand photoinduced electron transfer strategy. The probe shows approximately 100-fold increase of luminescence upon N-acetylation catalyzed by NAT, with relatively hi...

Epigenetic targets and drug discovery: part 1: histone methylation.

Dynamic chromatin structure is modulated by post-translational modifications on histones, such as acetylation, phosphorylation and methylation. Research on histone methylation has become the most flourishing area of epigenetics in the past fourteen years, and a large amount of data has been accumulated regarding its biology and disease implications. Correspondingly, a lot of efforts have been m...
